Overview

Prg25-02 is a preclinical-stage small molecule inhibitor being developed by Tessellate Bio in collaboration with Boehringer Ingelheim. It targets the Fanconi anemia group M protein (FANCM) complex, which is essential for the Alternative Lengthening of Telomeres (ALT) pathway. By inhibiting FANCM, Prg25-02 leverages synthetic lethality to selectively kill ALT-positive cancer cells—found in approximately 10-15% of cancers, including many sarcomas and brain tumors—while sparing healthy cells. The program includes the development of a companion C-Circle assay diagnostic to identify patients likely to benefit from the treatment.
