Overview

PRGN-2009 is a therapeutic cancer vaccine based on a gorilla adenovirus vector platform (AdenoVerse technology) developed to target cancers associated with human papillomavirus (HPV) types 16 and 18. It delivers HPV16 and HPV18 antigens designed to induce robust T cell immune responses against cancer cells expressing these viral proteins. The vaccine contains multiple cytotoxic T cell epitopes from the E6 and E7 oncoproteins of HPV16/18, aiming to stimulate both CD8+ and CD4+ T-cell responses. Preclinical studies have demonstrated its immunogenicity, and early-phase clinical trials have shown safety as well as induction of anti-HPV immune responses in patients with recurrent or metastatic HPV-positive malignancies. It is being evaluated alone or in combination with checkpoint inhibitors such as pembrolizumab for enhanced antitumor efficacy[1][5][6].
