Overview

PRI-002 is an investigational, orally available all-D-enantiomeric peptide developed for the treatment of early-stage Alzheimer's disease, including mild cognitive impairment (MCI) and mild dementia due to Alzheimer's. Its mechanism of action is unique among amyloid-targeting therapies: PRI-002 directly disassembles toxic amyloid-beta (Aβ) oligomers—considered key drivers of neurodegeneration in Alzheimer’s—into harmless monomers through a purely physical, chaperone-like process. This anti-prionic approach does not involve the immune system and distinguishes it from antibody-based drugs. Preclinical studies demonstrated reversal of cognitive deficits in multiple animal models, and Phase 1 trials established safety and tolerability in humans. A large Phase 2 trial (PRImus-AD) is ongoing to assess efficacy in patients with early Alzheimer’s disease[1][3][4][5][6][7][9].
