PRI-101 is an orally available all-d-peptide drug candidate developed by Priavoid GmbH using its proprietary 'detangler' platform. It is designed to treat synucleinopathies, such as Parkinson's disease, multiple system atrophy (MSA), and Lewy body dementia, by targeting and disassembling toxic α-synuclein (α-syn) aggregates. Unlike traditional approaches that may target monomeric α-synuclein, PRI-101 binds to α-synuclein subunits within existing aggregates and promotes their conversion back into non-toxic forms. Preclinical studies have demonstrated that PRI-101 can cross the blood-brain barrier, reduce pathological α-synuclein load, and improve survival and behavioral performance in animal models. The development of PRI-101 is supported by a grant from The Michael J. Fox Foundation for Parkinson's Research.
