Overview

pri-miR-126 is the primary microRNA precursor for miR-126 and miR-126(*). This microRNA pair has been shown to independently suppress the sequential recruitment of mesenchymal stem cells and inflammatory monocytes into the tumor stroma, thereby inhibiting lung metastasis by breast tumor cells in a mouse xenograft model. Its expression is downregulated in cancer cells due to promoter methylation of its host gene Egfl7, and this downregulation correlates with poor metastasis-free survival in breast cancer patients. The mechanism involves direct inhibition of stromal cell-derived factor-1 alpha (SDF-1α) expression and indirect suppression of chemokine (C-C motif) ligand 2 (Ccl2) expression by cancer cells in an SDF-1α-dependent manner.
