PRI5202 is a synthetic analog of 1,25-dihydroxyvitamin D3 (calcitriol) developed for the treatment of hematological malignancies. It acts as a differentiation-inducing agent, specifically targeting the Vitamin D receptor (VDR) to promote the maturation of leukemic blasts into more mature myeloid cells. Research indicates that PRI5202, particularly when combined with low-dose fludarabine, exhibits synergistic immunostimulatory and pro-differentiation effects in acute myeloid leukemia (AML) cells, especially those with FGFR and JAK pathway mutations. It has also shown potential in inducing myeloid differentiation in blasts from patients with myelodysplastic syndrome (MDS).
