Drug intelligence / Profile preview
Prilocaine hydrochloride is an intermediate-acting local anesthetic of the amide class, chemically related to lidocaine. It acts by binding to the intracellular portion of voltage-gated sodium channels in the neuronal membrane, preventing sodium influx during depolarization. This stabilizes the membrane and inhibits the initiation and conduction of nerve impulses, resulting in a reversible local anesthetic effect. Prilocaine is widely used for infiltration anesthesia, peripheral nerve blocks, and spinal/epidural anesthesia, particularly in dental and minor surgical procedures. It is characterized by a rapid onset and moderate duration of action, with lower cardiotoxicity compared to some other amide anesthetics, though its metabolism can lead to the formation of o-toluidine, which carries a risk of inducing methemoglobinemia.
