PRIME-2-CoV_Beta is a novel COVID-19 vaccine candidate based on an attenuated, second-generation Orf virus (ORFV) vector. It is engineered to express both the spike and nucleocapsid proteins of SARS-CoV-2, specifically including the receptor-binding domain (RBD) of the Beta variant. The vaccine aims to induce broad immune responses by presenting multiple viral antigens. In first-in-human phase I clinical trials, it was administered as a booster in healthy adults and demonstrated a favorable safety profile with only mild or moderate adverse events reported. Immunogenicity data showed robust antigen-specific humoral and cellular immune responses, including significant increases in neutralizing antibodies against SARS-CoV-2 and its variants. Notably, administration did not induce ORFV-neutralizing antibodies, suggesting potential for repeated dosing without vector immunity concerns[1][6][9].
