PRIMVAC is a recombinant protein-based vaccine candidate designed to prevent placental malaria, a severe complication of Plasmodium falciparum infection during pregnancy. The vaccine is based on the CSA-binding DBL1x-2x multidomain of the VAR2CSA protein, which mediates sequestration of infected erythrocytes in the placenta. By inducing antibodies against VAR2CSA, PRIMVAC aims to block this interaction and protect pregnant women from adverse outcomes associated with placental malaria. The vaccine has been shown in preclinical and phase 1 clinical studies to be safe, immunogenic, and capable of inducing functional antibodies that recognize homologous VAR2CSA variants[1][2][3][4][5]. It is administered intramuscularly and has been tested with adjuvants such as Alhydrogel or GLA-SE.
