Overview

Pritumumab is a natural human IgG1 kappa monoclonal antibody that targets the ecto-domain of vimentin (cell surface vimentin) expressed on various tumor cell types but not on normal tissues. It was originally derived from a B lymphocyte isolated from a regional draining lymph node of a patient with cervical carcinoma. Pritumumab is being developed primarily for the treatment of gliomas and other brain cancers by Nascent Biotech. Its mechanism involves binding to cell surface vimentin, leading to antibody-dependent cellular cytotoxicity and inhibition of tumor growth. The drug has also been proposed as an antiviral agent due to its target’s role in viral replication processes such as those seen in SARS-CoV and COVID-19[1][7][9]. Clinical trials have shown it to be well tolerated with no dose-limiting toxicities up to 16.2 mg/kg weekly[5][6][10]. Orphan drug designation has been granted for glioma.
