Pritumumab ADC is an antibody-drug conjugate (ADC) being developed by Manhattan BioSolutions in collaboration with Nascent Biotech. The candidate consists of pritumumab, a natural human monoclonal antibody that targets the extracellular form of vimentin, conjugated to a cytotoxic payload. Vimentin is an intermediate filament protein that is overexpressed on the surface of various cancer cells, particularly in brain and pancreatic cancers, and is associated with tumor growth and metastasis. The ADC utilizes Manhattan BioSolutions' proprietary platform, which typically employs a legumain-activated linker and a topoisomerase I inhibitor (TOP1i) payload, such as exatecan derivatives. Pritumumab has demonstrated the ability to cross the blood-brain barrier, potentially allowing the ADC to treat primary brain tumors and brain metastases. The program is currently in the preclinical stage, with studies focused on vimentin-positive advanced or metastatic solid tumors.
