Overview

PRMT4 shRNA is a short hairpin RNA designed to silence the expression of Protein Arginine Methyltransferase 4 (PRMT4), also known as Coactivator-Associated Arginine Methyltransferase 1 (CARM1). In the context of acute myeloid leukemia (AML), PRMT4 is frequently overexpressed and plays a critical role in maintaining the leukemic phenotype. By utilizing lentiviral delivery, PRMT4 shRNA effectively knocks down the target protein, leading to impaired cell cycle progression, reduced cell proliferation, and the induction of apoptosis in leukemia cell lines. Research indicates that PRMT4 knockdown specifically down-regulates E2F target genes and reduces H3R17 dimethylation at their promoters. This modality serves as a research tool to validate PRMT4 as a therapeutic target, demonstrating selective dependency in leukemia cells compared to normal hematopoietic stem and progenitor cells.
