Overview

PRN-001-03 is a small molecule drug candidate being developed by Proniras for the treatment of alcohol withdrawal. It is part of the PRN-001 series, which is based on tezampanel, a competitive antagonist of both α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) and kainate receptors. These receptors are subtypes of ionotropic glutamate receptors that mediate fast excitatory synaptic transmission in the central nervous system. During alcohol withdrawal, the brain experiences a state of glutamatergic hyperexcitability; PRN-001-03 is designed to counteract this by blocking the overactivation of AMPA and kainate receptors. As of early 2024, PRN-001-03 is in the preclinical stage of development.
