Drug intelligence / Profile preview
PRN2675 is a central nervous system (CNS)-penetrant, highly potent, selective, and irreversible covalent small molecule inhibitor of Bruton's tyrosine kinase (BTK). Developed by Principia Biopharma (later acquired by Sanofi), PRN2675 is a structural analogue of tolebrutinib, differing by a single fluorine substitution for hydrogen near the reactive moiety. It is widely used as a preclinical tool compound to evaluate the therapeutic potential of BTK inhibition in neuroinflammatory and demyelinating conditions, such as multiple sclerosis (MS). In preclinical models, including experimental autoimmune encephalomyelitis (EAE) and cuprizone-induced demyelination, PRN2675 has been shown to halt disease progression, reduce plasma neurofilament heavy chain (NfH) levels, and modulate the expression of genes associated with disease-associated microglia (DAM), thereby mitigating microglia-driven neuroinflammation.
