PRN694 is a potent, small-molecule, covalent inhibitor of the Tec family kinases ITK (Interleukin-2-inducible T-cell kinase) and RLK (Resting lymphocyte kinase, also known as TXK). Developed by Principia Biopharma (now part of Sanofi), PRN694 is designed to selectively and irreversibly block these kinases, which are primarily expressed in T cells and are critical components of the T-cell receptor (TCR) signaling pathway. By inhibiting ITK and RLK, PRN694 suppresses the activation, proliferation, and migration of T lymphocytes, as well as the production of proinflammatory cytokines such as IL-17A. Preclinical studies in mouse models of psoriasis have shown that PRN694 reduces epidermal thickness, hyperproliferation, and T-cell infiltration, suggesting its potential as a therapeutic agent for Th17-mediated inflammatory skin disorders.
