PRNP-targeted siRNA is an experimental small interfering RNA (siRNA) designed to silence the expression of the *PRNP* gene, which encodes the cellular prion protein (PrPc). In the context of triple-negative breast cancer (TNBC), PrPc has been identified as a promoter of tumor aggressiveness, facilitating migration, invasion, and proliferation. By transiently silencing *PRNP*, this siRNA attenuates the canonical WNT/β-catenin signaling pathway, leading to reduced β-catenin levels and a shift in epithelial-mesenchymal transition (EMT) markers, such as increased E-cadherin and decreased N-cadherin expression. This molecular intervention results in growth arrest in the G0 phase and reduced metabolic activity in cancer cells, suggesting that targeting PrPc may offer a promising therapeutic strategy to limit the aggressiveness of TNBC.
