PRO 2000

PRO 2000 is an investigational synthetic polyanionic compound developed as a topical vaginal microbicide for the prevention of sexually transmitted infections, primarily HIV. It is designed to be applied vaginally prior to sexual intercourse and acts by blocking the entry of pathogens such as HIV into human cells. Mechanistically, it binds to HIV gp120 and interferes with virus attachment and/or fusion with CD4+ T cells. Additionally, it interacts with CD4 and CXCR4 receptors on host cells and exhibits antagonistic activity at CXCR4, which may explain its greater activity against X4-tropic viruses compared to R5-tropic viruses. Laboratory studies have shown activity against other sexually transmitted pathogens including herpes simplex virus (HSV), chlamydia, and Neisseria gonorrhoeae. Despite promising preclinical results, large phase III clinical trials demonstrated that PRO 2000 was not effective in preventing HIV transmission in women[1][2][3][6].
