PRO CAR-202 is an autologous, IL-18 armored CAR T-cell therapy targeting Six-transmembrane epithelial antigen of the prostate 1 (STEAP1). Developed by PromiCell Therapeutics and exclusively licensed from the Fred Hutchinson Cancer Center, it is designed for the treatment of metastatic castration-resistant prostate cancer (mCRPC) and Ewing Sarcoma. The therapy utilizes a humanized single-chain variable fragment (scFv) with high affinity for STEAP1. The "armoring" component involves the expression of the pro-inflammatory cytokine IL-18, which is intended to modulate the tumor microenvironment, enhance T-cell persistence, and stimulate immune responses against both antigen-positive and antigen-negative tumor cells. PRO CAR-202 builds upon the PRO CAR-201 program and is currently in the IND-enabling phase, with clinical enrollment anticipated to begin in early 2026.
