Pro insulin peptide is a synthetic fragment of the human proinsulin protein, designed for use as an antigen-specific immunotherapy in type 1 diabetes. Proinsulin is the precursor to insulin and C-peptide, produced by pancreatic beta cells[1][2]. In clinical research, specific peptides derived from the proinsulin sequence are administered intradermally to modulate immune responses against pancreatic beta cell antigens. The goal is to induce immune tolerance and preserve residual beta cell function in newly diagnosed type 1 diabetes patients. Mechanistically, these peptides are thought to promote regulatory T cell responses (e.g., increased FoxP3 expression), stimulate interleukin-10 production upon antigen challenge, and reduce pathogenic T cell activity targeting beta cells[6][7]. Early-phase clinical trials have shown that this therapy can be safe and may help maintain endogenous insulin production without accelerating disease progression[6][7].
