PRO1102 is a novel, preclinical-stage antibody-drug conjugate (ADC) targeting human epidermal growth factor receptor 2 (HER2). Developed by ProfoundBio (which was acquired by Genmab in 2024), PRO1102 is composed of the clinically validated anti-HER2 monoclonal antibody trastuzumab conjugated to exatecan, a topoisomerase 1 inhibitor payload, via a novel cleavable, hydrophilic linker. In preclinical models, PRO1102 demonstrated robust anti-tumor activity across multiple HER2-expressing cell lines, including those with moderate and low HER2 expression, showing superior potency compared to ADCs utilizing alternative drug-linker technologies (such as MMAE, DM1, and deruxtecan). It has shown stable pharmacokinetics in rats and a favorable tolerability profile in mice.
