PRO1107 is an investigational antibody-drug conjugate (ADC) targeting Protein Tyrosine Kinase 7 (PTK7), developed for the treatment of advanced solid tumors. The ADC consists of a PTK7-targeted monoclonal antibody conjugated to a novel, proprietary hydrophilic monomethyl auristatin E (MMAE)-based linker-drug platform (LD343) at a high drug-antibody ratio of 8. MMAE is a potent microtubule inhibitor that disrupts cell division and induces apoptosis in cancer cells. The hydrophilic linker technology enables efficient delivery and improved pharmacokinetics by masking the hydrophobicity of MMAE, allowing higher payload loading while maintaining favorable properties. Preclinical data suggest superior antitumor activity and tolerability compared to prior PTK7-directed ADCs, attributed to enhanced tumor delivery and reduced systemic exposure to free payload. PRO1107 is currently being evaluated in global Phase 1/2 clinical trials for multiple cancers expressing PTK7, including non-small cell lung cancer, breast cancer (including triple negative), ovarian cancer, endometrial carcinoma, gastroesophageal junction adenocarcinoma, urothelial carcinoma, and others[3][6][8][9].
