PRO1160

PRO1160 is a novel antibody-drug conjugate (ADC) designed to target CD70, an antigen that mediates immunosuppression and is overexpressed in multiple solid tumors and hematologic malignancies, with limited expression in normal tissues. The drug consists of a human monoclonal antibody specific for CD70, a protease-cleavable proprietary hydrophilic linker, and exatecan—a topoisomerase 1 inhibitor—as the cytotoxic payload. The hydrophilic linker improves physicochemical properties and pharmacokinetics compared to conventional linkers. Exatecan is membrane permeable, not a substrate for multidrug resistance pumps, and provides strong bystander effects. Preclinical studies have shown high potency against renal cell carcinoma (RCC), non-Hodgkin lymphoma (NHL), and nasopharyngeal carcinoma (NPC). PRO1160 is currently being evaluated in an open-label Phase 1/2 clinical trial for patients with metastatic RCC, metastatic or relapsed NPC, or advanced relapsed/refractory NHL[1][2][3][5][7].
