Overview

ProAgio is a pegylated peptide drug developed from a human protein that targets and kills cells expressing integrin αvβ3, such as cancer-associated fibroblasts (CAFs) and endothelial cells. By binding to the αvβ3 integrin receptor, which is present on abnormal endothelial cells forming tumor blood vessels and on CAFs that generate the physical barrier around tumors, ProAgio induces apoptosis in these cancer-supporting cells. This mechanism disrupts the dense stroma of solid tumors—such as pancreatic cancer and triple-negative breast cancer—improving drug delivery to tumor sites by reopening collapsed blood vessels and reducing irregular angiogenic vasculature. Preclinical studies have shown efficacy in murine models of pancreatic and breast cancers, with ongoing clinical trials evaluating its safety and recommended dosing in humans[1][4][5][6][7].
