Probody T-cell bispecifics (TCBs) are a class of conditionally activated, bispecific T-cell engaging antibodies developed by CytomX Therapeutics in collaboration with partners such as Regeneron and Bristol Myers Squibb. These molecules are engineered using CytomX's proprietary Probody technology, which involves masking the antibody's binding domains with a peptide linker that is sensitive to tumor-associated proteases. This design is intended to restrict T-cell activation and tumor-cell killing to the tumor microenvironment, thereby reducing the systemic 'off-tumor' toxicities often associated with conventional T-cell engagers. While specific tumor-associated antigens for these candidates remain undisclosed, the molecules are designed to simultaneously bind a tumor antigen and the CD3 receptor on T cells to facilitate immune-mediated tumor destruction. They are currently in preclinical development for the treatment of various solid tumors.
