Procarbazine + lomustine + vincristine (commonly referred to as "PCV") is a combination chemotherapy regimen used primarily in the treatment of certain brain tumors, especially gliomas and anaplastic oligodendroglial tumors. Each component has a distinct mechanism of action: - Procarbazine is an alkylating agent that inhibits DNA, RNA, and protein synthesis by causing DNA strand breaks. - Lomustine (also known as CCNU) is a nitrosourea alkylating agent that cross-links DNA and RNA. - Vincristine is a vinca alkaloid that inhibits microtubule formation in mitotic spindle fibers during cell division. Together, these drugs target rapidly dividing cancer cells through complementary mechanisms. The regimen has been shown to improve overall survival when combined with radiation therapy for certain gliomas[1][5]. Administration involves oral capsules for procarbazine and lomustine and intravenous infusion for vincristine[1][2].
