Overview

Prochlorperazine is a first-generation (typical) antipsychotic of the phenothiazine class. It is primarily used to treat severe nausea and vomiting, schizophrenia, and non-psychotic anxiety. Prochlorperazine acts mainly as a dopamine D2 receptor antagonist in the brain, which reduces dopaminergic neurotransmission responsible for psychotic symptoms and triggers for vomiting. It also exhibits antagonistic activity at histaminergic, cholinergic (muscarinic), and alpha-1 adrenergic receptors. Compared to other phenothiazines, it is less sedating and has a lower risk of causing hypotension or potentiating CNS depressants[4][5][6]. The drug is available in oral tablets (including dissolvable forms), rectal suppositories, and injectable formulations[4][8].
