Overview

ProCP-2 is a recombinant human pro-carboxypeptidase B2 (pro-CPB2), also known as pro-thrombin-activatable fibrinolysis inhibitor (pro-TAFI), currently under development by Prothix. As a zymogen, ProCP-2 is activated by the thrombin-thrombomodulin complex into TAFIa, a basic carboxypeptidase that regulates both the coagulation and complement systems. TAFIa stabilizes blood clots by removing C-terminal lysine residues from fibrin, which prevents the binding and activation of plasminogen and tissue plasminogen activator (tPA), thereby slowing fibrinolysis. Beyond its role in hemostasis, TAFIa exerts potent anti-inflammatory effects by enzymatically inactivating pro-inflammatory peptides, including the complement anaphylatoxins C3a and C5a, bradykinin, and osteopontin. ProCP-2 is being evaluated in clinical trials for its potential to treat acute inflammatory and thrombotic conditions, such as sepsis, where dysregulated fibrinolysis and 'complement storm' contribute to organ failure and mortality.
