Prodrug Supercarriers are a nanoparticle-based delivery system developed by Temprian Oncology (a subsidiary of Temprian Therapeutics) for the treatment of metastatic melanoma. Developed in collaboration with Northwestern University's Le Poole and Nguyen Laboratories, these "Supercarriers" utilize MBEH (monobenzone) nanoparticle technology to encapsulate a cytotoxic payload. The delivery vehicle is engineered to be stable in systemic circulation but undergoes a conformational change to release its active cytotoxic ingredient specifically within the low-pH environment of melanosomes—specialized organelles in melanoma cells. This targeted approach aims to maximize therapeutic efficacy against tumor cells while minimizing off-target systemic toxicity.
