Overview
**Profavax** is a live-attenuated herpes simplex virus type 2 (HSV-2) prophylactic vaccine developed by Rational Vaccines (RVx) to prevent genital herpes infection caused by HSV-2. It mimics the structure of the wild-type virus and, in preclinical animal models (mice and guinea pigs), demonstrated approximately 100 times greater efficacy than subunit vaccines like GEN-003 or glycoprotein D-based candidates, which have repeatedly failed in clinical trials. The vaccine aims to induce robust protective immunity against initial HSV-2 infection, with plans for Phase I safety trials estimated at 2-3 years from 2016, though development was led by chief science officer William Halford, who passed away in 2017, potentially impacting progress.[1][5][7]
