Profenamine (also known as ethopropazine) is a phenothiazine derivative used primarily as an antiparkinsonian agent and antidyskinetic. It acts mainly through anticholinergic (antimuscarinic) activity by partially blocking central striatal cholinergic receptors to help balance cholinergic and dopaminergic activity in the basal ganglia. This mechanism alleviates symptoms such as rigidity, akinesia, sialorrhea, oculogyric crisis, and tremor associated with Parkinson's disease of various origins (encephalitic, arteriosclerotic or idiopathic), as well as drug-induced extrapyramidal reactions. Profenamine also exhibits antihistaminic and antiadrenergic properties but is not effective against tardive dyskinesia[1][2][4][5].
