Progabide is a small molecule, fatty acid derivative and an analog and prodrug of gamma-aminobutyric acid (GABA), primarily used in the treatment of epilepsy. It acts as an agonist at GABAA, GABAB, and GABAA-rho receptors, enhancing inhibitory neurotransmission in the central nervous system. By binding to these receptors on primary afferent fibers, progabide increases chloride ion flux (via GABAA) and reduces calcium influx (via GABAB), leading to increased presynaptic inhibition and reduced release of excitatory neurotransmitters. Progabide has also been investigated for use in Parkinson's disease, schizophrenia, clinical depression, anxiety disorders, neuroleptic-induced dyskinesia, L-dopa-induced involuntary movements, and mania; however its efficacy for these indications remains unclear[1][2][3][4][6].
