Overview

Proglumetacin is a nonsteroidal anti-inflammatory drug (NSAID) of the indole acetic acid derivative class. It is primarily used to manage pain and inflammation associated with musculoskeletal and joint disorders such as rheumatoid arthritis and osteoarthritis. Proglumetacin acts as a prodrug that is metabolized in the body to indometacin (an active NSAID) and proglumide (which has antisecretory effects that help protect the gastric mucosa). Its mechanism of action involves inhibition of cyclooxygenase enzymes at sites of inflammation—thereby reducing prostaglandin synthesis—and it may also inhibit 5-lipoxygenase activity. The drug was first developed by Fresenius[1][2][3][4][5][6][7][8].
