Program 3 is a preclinical small molecule program being developed by Atavistik Bio, focused on a mutant-selective allosteric inhibitor of JAK2 V617F for the treatment of myeloproliferative neoplasms (MPNs). Developed using the company's proprietary AMPS (Atavistik Metabolite-Protein Screening) platform, these inhibitors bind to the allosteric JH2 pseudokinase domain, the specific site of the V617F mutation. This mechanism allows for potent inhibition of mutant JAK2-dependent signaling and cell proliferation while sparing wild-type JAK2 cells, potentially offering a better safety profile and broader therapeutic window than current orthosteric JAK inhibitors. The program has demonstrated picomolar affinity for the JAK2 V617F JH2 domain and high selectivity over other JAK family kinases in preclinical models.
