Programmed cell death protein 1 knockout engineered T cells are an investigational autologous cell therapy developed by Anhui Province Hospital. The therapy involves the ex vivo modification of a patient's own T cells using CRISPR/Cas9 gene-editing technology to permanently disrupt the PDCD1 gene, which encodes the PD-1 protein. PD-1 is a critical immune checkpoint receptor that, when activated by its ligands PD-L1 or PD-L2, suppresses T-cell effector functions and promotes immune evasion by tumor cells. By eliminating PD-1 expression, these engineered T cells are designed to resist exhaustion and maintain potent cytotoxic activity within the immunosuppressive tumor microenvironment. This candidate is currently being evaluated in Phase 1 clinical trials for the treatment of advanced solid tumors, including metastatic bladder cancer, renal cell carcinoma, and prostate cancer.
