Overview

Programmed cell death protein 1 (PD-1) knockout T cells are an autologous cell therapy developed by Majesale (Beijing) Biotechnology Co., Ltd., primarily for the treatment of non-small cell lung cancer (NSCLC). This therapy involves extracting T cells from a patient and using CRISPR/Cas9 gene-editing technology to disrupt the PDCD1 gene, which encodes the PD-1 checkpoint protein. By removing PD-1, the T cells are rendered resistant to PD-L1-mediated immunosuppression within the tumor microenvironment, thereby enhancing their anti-tumor activity upon re-infusion into the patient. This approach represents one of the first clinical applications of CRISPR technology in human oncology, aiming to overcome the limitations of systemic checkpoint inhibitors by providing a more targeted and durable immune response.
