A **programmed death-ligand 2 antibody** is an experimental monoclonal antibody or biologic therapeutic designed to specifically bind to **programmed death-ligand 2 (PD-L2)**, a cell surface protein that is a key ligand for the programmed death-1 (PD-1) immune checkpoint receptor. PD-L2 is expressed primarily on antigen-presenting cells and certain tumor cells and is implicated in the suppression of T cell activation and function, allowing tumors to evade immune detection. By blocking PD-L2 (alone or in concert with PD-L1), these antibodies aim to release the brakes on the immune system and enhance antitumor immune responses. This therapeutic strategy represents an emerging immune checkpoint blockade approach distinct from, but related to, anti-PD-1 and anti-PD-L1 therapies. As of 2025, anti-PD-L2 antibodies are investigational agents and have not yet entered routine clinical use. There are no approved, named monoclonal antibodies specific for PD-L2 in routine clinical use, and preclinical/early phase clinical research is ongoing[1][2][4][7][9].
