ProHema is an ex vivo pharmacologically-modulated hematopoietic stem cell (HSC) therapy developed by Fate Therapeutics. It is produced through a proprietary two-hour process where a single unit of umbilical cord blood is treated with 16,16-dimethyl prostaglandin E2 (dmPGE2), a stable analog of prostaglandin E2. This modulation is designed to activate key biological pathways involved in HSC homing, survival, and proliferation. ProHema was primarily developed for use in patients with hematologic malignancies undergoing double umbilical cord blood transplantation (dUCBT) to accelerate neutrophil recovery and enhance engraftment. Clinical development reached Phase 2 for hematologic malignancies and was also explored in pediatric patients with inherited metabolic disorders before Fate Therapeutics shifted its strategic focus toward induced pluripotent stem cell (iPSC)-derived therapies.
