Prohibitin binding peptide (PBP) is a therapeutic peptide that targets prohibitin (PHB), a protein involved in mitochondrial function and cell signaling that is often upregulated in cancer and adipose tissue. PBP acts by inhibiting glucose metabolism in tumor cells and reducing the levels of circulating leptin, an adipokine that contributes to PD-1-mediated immune cell exhaustion in obese environments. By modulating these metabolic and endocrine pathways, PBP helps convert the tumor microenvironment from an immunosuppressive to an immunostimulatory state, thereby sensitizing tumors to PD-1 checkpoint inhibition. It is currently being researched as a potential treatment for melanoma, particularly in patients with obesity-associated chronic inflammation.
