Overview

Proinsulin II-GAD mRNA lipid particle aggregates is an experimental immunotherapeutic vaccine candidate designed for the prevention or delay of type 1 diabetes (T1D). It consists of mRNA encoding two key beta-cell autoantigens, proinsulin II (PI2) and glutamic acid decarboxylase (GAD), encapsulated within anionic lipid particle aggregates (LPAs). The mechanism involves the delivery of these autoantigens to the immune system in a manner that promotes immune tolerance and suppresses the autoimmune destruction of insulin-producing beta cells. In preclinical studies using non-obese diabetic (NOD) mice, the combination of PI2 and GAD mRNA-LPAs demonstrated superior efficacy in delaying the onset of diabetes compared to single-antigen formulations, particularly at the 18-week mark.
