Prokaryotics MsbA inhibitor is a preclinical-stage small molecule antibiotic being developed by Prokaryotics, Inc. for the treatment of complicated urinary tract infections (cUTI) caused by Enterobacteriaceae. The drug targets MsbA, an essential ATP-binding cassette (ABC) transporter that functions as a lipid A flippase, moving lipopolysaccharide (LPS) from the inner leaflet of the cytoplasmic membrane to the periplasmic leaflet. By inhibiting this transport process, the drug disrupts the biogenesis of the bacterial outer membrane, leading to a loss of cell viability. The program is being optimized for both intravenous (IV) and oral administration and has received significant funding from the NIH (grant 1R01 AI165740) to support lead selection and preclinical development.
