Overview

Prolanta is a recombinant human prolactin (hPRL) analog that acts as a potent and selective **prolactin receptor (PRLR) antagonist**. Developed by Oncolix, Inc., the drug is a modified version of the natural prolactin hormone containing a single amino acid substitution (glycine to arginine at position 129). This mutation allows the molecule to bind to the prolactin receptor with high affinity but prevents the conformational change and subsequent receptor dimerization necessary for signal transduction. By blocking the PRLR, Prolanta inhibits the activation of the **JAK2/STAT5** and **MAPK** signaling pathways, which are frequently exploited by certain cancers to promote cell proliferation, survival, and resistance to chemotherapy. Prolanta has been primarily investigated in Phase I clinical trials for the treatment of recurrent or persistent epithelial ovarian, primary peritoneal, and fallopian tube cancers, where the prolactin receptor is often overexpressed.
