Drug intelligence / Profile preview
Prolease-r-hFSH is a recombinant human follicle stimulating hormone (r-hFSH) formulated with Alkermes' ProLease injectable sustained release drug delivery technology[6][8]. This long-acting formulation is designed to provide patients with a single injection alternative to multiple daily injections for the treatment of infertility[2][4]. The drug was developed through a collaboration between Alkermes, Inc. and Serono S.A., which was announced in January 2000[2]. According to the search results, the drug completed a Phase I clinical trial, and further clinical studies were planned as of 2002[4]. The decision to proceed with clinical development followed the successful completion and analysis of data from a Phase I trial, which triggered an undisclosed milestone payment to Alkermes[2]. The ProLease technology used in this formulation is based on the encapsulation of drugs into small polymeric microspheres that degrade slowly and release drugs at a controlled rate following subcutaneous or intramuscular injection[7]. This approach is particularly valuable for protecting fragile molecules from degradation during the manufacturing process[7]. As a recombinant form of follicle stimulating hormone, Prolease-r-hFSH works by binding to the follicle stimulating hormone receptor, which is a G-coupled transmembrane receptor[3]. This binding appears to induce phosphorylation and activation of the PI3K and Akt signaling pathway, which regulates many metabolic and related survival/maturation functions in cells[3]. The primary application of Prolease-r-hFSH is in the treatment of infertility[1][2]. Specifically, follitropin (the active component) is used to: * Stimulate follicle development in women with primary ovarian hypogonadism * Treat female infertility by stimulating ovarian follicular growth in women without primary ovarian failure * Induce spermatogenesis in men with hypogonadotropic hypogonadism where infertility is not due to primary testicular failure[3][5] The collaboration between Alkermes and Serono was significant, with Serono providing development funding and milestone payments that could exceed $25 million, plus royalties on sales of any resulting ProLease products[2].
