PR-HMRG is a novel, spray-type fluorescent probe developed for the intraoperative rapid detection of malignant glioma, specifically glioblastoma. It consists of a hydroxymethyl rhodamine green (HMRG) fluorescent scaffold conjugated to a proline-arginine dipeptide. The probe is designed to be enzymatically activated by Calpain-1, a protease that is significantly overexpressed in glioma tumor tissues. Upon cleavage of the dipeptide by Calpain-1, the HMRG scaffold becomes highly fluorescent, allowing for real-time visualization of tumor margins during surgery. This probe aims to address limitations of current surgical adjuncts like 5-aminolevulinic acid (5-ALA), such as false results and the inability to re-administer the agent during a single procedure.
