Drug intelligence / Profile preview
Promethazine teoclate is a long-acting, first-generation antihistamine and antiemetic. It is a phenothiazine derivative formulated as a salt with 8-chlorotheophylline (theoclate) to provide a prolonged duration of action (up to 24 hours) compared to the hydrochloride salt. Promethazine teoclate acts primarily as an antagonist at histamine H1 and muscarinic acetylcholine receptors, which underlies its efficacy in preventing and treating motion sickness, vertigo, labyrinthitis, Ménière's disease, and postoperative nausea and vomiting. It also exhibits antagonist activity at dopamine D2, alpha-1 adrenergic, 5-HT2, and NMDA receptors, contributing to its sedative properties. While historically developed by Rhône-Poulenc, it is widely available as a generic medication and is currently listed under development as an active pharmaceutical ingredient (API) for psychosis by Bajaj Healthcare.
