Promiximab is a novel humanized IgG1 monoclonal antibody targeting CD56 (Neural Cell Adhesion Molecule, NCAM), specifically developed for the treatment of small cell lung cancer (SCLC). It exhibits high affinity and efficient internalization upon binding to CD56-expressing cells. Promiximab serves as the targeting component of the antibody-drug conjugate (ADC) promiximab-DUBA, which utilizes a duocarmycin-based DNA-alkylating payload. In preclinical studies, this ADC has shown potent in vitro cytotoxicity and significant in vivo antitumor efficacy, including sustained tumor regression in SCLC xenograft models without significant systemic toxicity.
