proNGF (pro-Nerve Growth Factor) is the precursor form of nerve growth factor (NGF), a neurotrophin involved in neuronal development, survival, and differentiation. Unlike mature NGF, proNGF can act as a signaling ligand with distinct biological activities. It binds preferentially to the p75 neurotrophin receptor (p75NTR), often in complex with sortilin, and can induce apoptosis or promote inflammation depending on cellular context. Recombinant forms of proNGF have been developed for research and potential therapeutic applications. In disease states such as brain injury or chronic inflammation, elevated levels of proNGF are associated with increased neuronal death and inflammatory cytokine production. The balance between proNGF and mature NGF is critical for modulating neuronal fate; dysregulation has been implicated in neurodegenerative diseases like Alzheimer’s disease, as well as chronic inflammatory conditions such as arthritis[1][2][4][5][7][9].
