Overview

Propargylglycine is a small molecule, non-proteinogenic amino acid and mechanism-based ("suicide") inhibitor of cystathionine gamma-lyase (CTH) and closely related enzymes, including proline dehydrogenase (PRODH/PRODH1) and 4-hydroxyproline dehydrogenase (PRODH2/HYPDH). The compound irreversibly inhibits cystathionine gamma-lyase, thereby blocking endogenous hydrogen sulfide (H2S) production, and can also target PRODHs, leading to anticancer activity via modulation of mitochondrial metabolism and stress response. It is under investigation for rare metabolic diseases such as primary hyperoxaluria, as well as potential repurposing for neurodegenerative disease mitigation via effects on neural mitochondrial function and mitohormesis. Propargylglycine is widely used in research on sulfur metabolism, neuroprotection, and enzyme affinity labeling[1][2][3][4][5][7][10].
