Overview
Prophylactic donor lymphocyte infusion (DLI) is an adoptive cellular immunotherapy protocol developed and evaluated by the Institut Paoli-Calmettes for patients with acute myeloid leukemia (AML) who have undergone allogeneic hematopoietic stem cell transplantation (Allo-HSCT). The procedure involves the infusion of unmanipulated lymphocytes, primarily T-cells, collected from the original stem cell donor. The therapeutic goal is to enhance the graft-versus-leukemia (GVL) effect, wherein the donor's immune cells recognize and destroy residual recipient leukemic cells that survived the transplant conditioning regimen. This approach is specifically investigated in Phase II trials as a preventative measure against disease relapse in high-risk AML populations, such as those with unfavorable cytogenetics or refractory disease.
