Propiin (S-propyl-L-cysteine sulfoxide) is a naturally occurring sulfur-containing amino acid derivative found in plants of the *Allium* genus, such as onions and garlic. In the field of oncology, it is being investigated as a prodrug substrate in a targeted enzyme prodrug therapy (TEPT) system. This therapeutic strategy utilizes a pharmacological pair consisting of propiin and a tumor-targeted enzyme, specifically a daidzein-conjugated recombinant methionine γ-lyase (C115H MGL-Dz). When propiin is processed by this enzyme, it is converted in situ into dipropyl thiosulfinate, a cytotoxic agent that induces oxidative stress and apoptosis in tumor cells. Preclinical studies have demonstrated that the application of this pharmacological pair significantly reduces tumor volume in xenograft models of colorectal, pancreatic, and prostate cancers, suggesting its potential as a novel strategy for treating solid tumors.
