**propofol + HSK3486** is a combination of two intravenous anesthetic agents used in clinical research for the induction and maintenance of general anesthesia and procedural sedation. **Propofol** is a well-established sedative-hypnotic that acts rapidly via potentiation of the gamma-aminobutyric acid type A (GABA~A~) receptor, resulting in CNS depression. **HSK3486** (ciprofol) is a novel 2,6-disubstituted phenol derivative and a potent propofol analogue, which similarly acts as a positive allosteric modulator and direct agonist of GABA~A~ receptors, especially the α1β2γ2 subtype. HSK3486 offers comparable anesthetic efficacy to propofol, with reduced incidence of injection-site pain and a higher therapeutic index. Research shows similar onset and duration of anesthesia, with HSK3486 generally requiring lower doses due to greater potency. Both drugs are metabolized rapidly, primarily via hepatic CYP450 enzymes. Combination use may be in the context of comparative phase II/III clinical trials for anesthetic efficacy and safety.
